Production of Virus - Free Microbulbs of Garlic (Allium sativum L.) by in Vitro Culture of Vegetative and Floral Buds in Immature Involucre

Abstract
By in vitro culture of vegetative and floral buds of garlic in a fast growing period, it was possible to produce virus-free stocks on a large scale which were not able to be routinely obtained by the conventional method of shoot meristem culture. As the electron microscopic examination revealed that a considerable amount of viruses existed on the shoot meristem per se of the bulb, it was found that the conventional method was not applicable to the production of virus-free stocks. During a fast growing period when the flower stem measured less than 5 §¯ in length, the apex of the stem had viruses at lower frequency than that of the bulb meristems. When the flower stems measured over 5 §¯, just before the involucer was protruded from the flag leaf, the number of virus-infected apices decreased further. However, all of the mature ¢¥bulbils¢¥ contained viruses. Seventy percent of the vegetative and floral buds in the immature involucer were free from virus contamination. The apex of flower stem gave rise to about 20 microbulbs when cultured in vitro. The results indicate that the number of viruses on the apex of the flower stem declines as the length of flower stem increases until the involucer was protruded from the flag leaf and that as the growth rate slows down, the movement of viruses seemingly catch up with the rate, resulting in virus infection of the apex.
